摘 要:目的研究鳖甲煎丸是否有治疗肝炎和抗肝纤维化的作用。方法采用刀豆素A(ConA)反复诱导的小鼠肝炎所致的肝纤维化模型,1.5、3.0、6.0 g/kg的鳖甲煎丸连续经口灌胃给药4周后取血检测血清肝功能指标[谷丙转氨酶(ALT)、谷草转氨酶(AST)、碱性磷酸酶(ALP)、总胆红素(TBIL)、白蛋白(ALB)]和炎性细胞因子[肿瘤坏死因子-α(TNF-α)、γ-干扰素(IFN-γ)、白细胞介素1β(IL-1β)],肝匀浆采用Elisa法检测肝纤维化指标[Ⅰ型胶原(CoⅠ),Ⅲ型胶原(CoⅢ)、Ⅳ型胶原(CoⅣ)、透明质酸(HA)、血清Ⅲ型前胶原(PCⅢ)、层黏蛋白(LN)],肝组织石蜡切片HE染色及Masson Trichrome染色观察肝脏结构及胶原纤维产生情况。结果各组小鼠间体质量无明显差异,模型组、阳性对照组及鳖甲煎丸给药组肝脏系数均高于阴性对照组(P<0.01);鳖甲煎丸对血清肝功能指标没有明显影响;3.0 g/kg和6.0 g/kg的鳖甲煎丸均降低了血清3个炎性细胞因子的水平,尤以TNF-α和IFN-γ明显(P值小于0.01或0.05);与模型组比较,3.0 g/kg和6.0 g/kg的鳖甲煎丸均明显降低了肝脏CoⅠ、CoⅢ、CoⅣ、PCⅢ、HA和LN的水平(P<0.01和0.05);Masson及HE染色结果显示鳖甲煎丸的3个剂量组均不同程度的使肝脏的纤维化组织减少或消失,使近半数肝脏结构恢复正常。结论鳖甲煎丸明显改善了由ConA诱导的小鼠肝炎和肝纤维化,具有显著的保肝和抗肝纤维化的作用。AIM To investigate the protective effect of Biejiajian Pills on hepatitis and hepatic fibrosis induced by ConA in mice. METHODS Mouse models of hepatic fibrosis were established by repeatedly injection of Co- nA. Biejiajian Pills were used for the prevention and treatment of hepatic fibrosis. The levels of alanine transami- nase (ALT), aspartate transaminase (AST), alkaline phosphatase (ALP), total bilirubin (TBIL), albumin (ALB) , tumor necrosis factor-α (TNF-α) , intefferon-γ (IFN-γ) and interleukin-1 β ( IL-1β ) in blood serum and procollagen Ⅲ ( PC Ⅲ ) , collagen I ( Co I ), collagen m ( Co Ⅲ ), collagen Ⅳ ( CoⅣ ), hyaluronic acid (HA) and laminin (LN) in liver homogenate were examined. Histological observation of the liver tissue was detected by Masson staining and HE stain. RESULTS The liver weight index in model group, colchicina group and Biejiajian Pills groups were significantly higher than that in the normal contrast group (P 〈 0. 01 ). Biejiajian Pills had no ob- vious effect on ALT, AST, ALP, TBIL and ALB in blood serum. Compared with model group, 6. 0 g/kg Biejiajian Pills significantly decreased the contents of TNF-α and IFN-γ (P 〈 0.01 or 0. 05 ). 3.0 g/kg and 6.0 g/kg Biejia- jian Pills significantly decreased the contents of Co I , Corn, ColV, Pcm, HA and LN in liver homogenate (P 〈 0. 01 or 0. 05 ). The degree and score of hepatic fibrosis staging were significantly lower in Biejiajian Pills groups than those in the model group (P 〈 0.01 ). CONCLUSION The experimental results of Biejiajian Pills show ob- vious protective effect on the hepatic fibrosis induced by ConA in mice.
