摘 要:目的研究补骨脂酚大鼠在体肠吸收动力学特征。方法应用大鼠在体单向灌流技术,同时建立HPLC法测定灌流液中补骨脂酚在大鼠不同肠段以及在不同质量浓度下的吸收情况。结果补骨脂酚在各肠段表观渗透系数均大于1.2×10-3cm/min,空肠的吸收速率与其他肠段相比有显著差异(P<0.05),随着补骨脂酚质量浓度增加,其在空肠的吸收速率常数(K a)和表观渗透系数(P app)值呈下降趋势。结论补骨脂酚在被考察的各个肠段吸收完全,最佳吸收部位是空肠;补骨脂酚的吸收中有高浓度饱和现象,可能存在主动转运或促进扩散。AIM To investigate the intestinal absorption kinetics of bakuchiol in rats. METHODS A single pass intestinal peffusion (SPIP) technique was used to measure the intestinal absorption of bakuchiol in different absorption segments and bakuchiol content was assayed by HPLC. The effect of mass concentrations on intestinal absorption was detected. RESULTS The apparent absorption coefficients of bakuehiol were more than 1.2 ×10 ^-3cm/min in each intestinal segment. HPLC data showed a higher absorption of bakuchiol in jejunum as compared with other intestinal segments (P 〈 0. 05 ). The absorption rate constant and apparent absorption coefficient of bakuehiol decreased in jejunum with the increase of mass concentration. CONCLUSION Bakuchiol is completely absorbed at all intestine segments in rats with the higher absorption rate in jejunum, and saturation phenomenon at high concentration may be relative to active transport or facilitated diffusion.
