高效液相色谱法测定天然生育酚中生育酚同系物含量的研究

摘　　要：建立了HPLC测定天然生育酚中各生育酚同系物含量的方法,试验采用ZorBax-C18(250 mm×4.6 mm,5μm)色谱柱,柱温为293.15 K,色谱级无水甲醇作为流动相,流速为1 mL/min,进样量为20μL,通过DAD检测器(波长为295 nm)测定生育酚异构体。结果表明:各生育酚峰面积与浓度线性关系良好,相关系数高;测得的天然生育酚样品中α-生育酚含量4.15%、β和γ-生育酚含量合为32.60%、δ-生育酚含量15.15%。本方法操作简便、重复性好,适合用于天然生育酚中各生育酚同系物含量的测定。A HPLC method for determination of each homolog of tocopherol in natural to-copherol has been established. A ZorBax-Cl8 （250 mm ×4.6 mm, 5 m） column was used as sta-tionary phase; The mobile phase was composed of methanol without water; The flow rate was 1.0 mL/min; The column temperature was 293. 15 K; The inject volume was 20 μL ;The to-copherol isomers were determined by DAD at Em 295 nm; The experiment results show that the peak area of each tocopherol keeps good linear relationship and high correlation coefficient with concentration; The composition of alpha-tocopherol, beta and gama-tocopherol, delta-tocopherol in natural tocopherol respectively is 4. 15%, 32.60% and 15.15%. This method is accurate and well replicability. It can be used in determination of each homolog of tocopherol in natural to-copherol.
