犀角地黄汤对胶原诱导性关节炎大鼠模型滑膜组织中磷脂酶C-γ1表达的影响

摘  要:目的探讨大鼠胶原诱导性关节炎(CIA)模型形成及应用犀角地黄汤治疗过程中大鼠关节滑膜组织中磷脂酶C-γ1(PLC-γ1)的表达情况,进一步探讨犀角地黄汤治疗类风湿关节炎的作用机制。方法 SD大鼠构建CIA模型,运用实时荧光定量反转录酶-聚合酶链锁反应(RT-PCR)法检测空白对照组、模型组和实验组大鼠在第42天关节滑膜组织中PLC-γ1的表达情况。结果模型组大鼠关节滑膜组织中PLC-γ1的相对表达量(0.125±0.001)明显高于空白对照组(0.031±0.001)(P<0.01),犀角地黄汤实验组PLC-γ1的相对表达量(0.072±0.001)低于模型组(P<0.05)。结论犀角地黄汤可降低大鼠关节滑膜组织中PLC-γ1的表达,对类风湿关节炎急性期有一定的治疗作用。.Objective To investigate the formation of collagen induced arthritis rat model and the influence of Comus Rhinoceri and Rehmanniae Decoction on the expression of phospholipase C-γ1 in rat arthrosis synovial tissue for rheumatoid arthritis treatment, in order to discuss Comus Rhinoeeri and Rehmanniae Decoction action mechanisms for rheumatoid arthritis treatment. Methods SD rats were used to build the animal model of collagen induced arthritis, the expression of phospholipase C-γ1 in arthrosis synovial tissues in control group, model group and experimental group were detected by real time fluores- cence quantitative polymerase chain reaction (FQ-PCR) on the 42th day. Results The relative expression of PLC-γ1 in arthro- sis synovial tissues of model group (0.125±0.001) was significantly higher than that of the control group (0.031±0.001) (P 〈 0.01); and the relative expression of PLC-γ1 in arthrosis synovial tissues of experimental group (0.072±0.001) was much lower than that of the model group (P 〈 0.05). Conclusion Comas Rhinoceri and Rehmannia Decoctioncan can lower the expressing of PLC-γ1 in arthrosis synovial tissues of rat, and has good therapeutic effect on acute phase of rheumatoid arthritis.
