摘 要:目的探讨特发性血小板减少性紫癜(ITP)模型小鼠体内,不同浓度的硝普钠(SNP)对活性氧(ROS)生成的影响及意义。方法 ITP模型小鼠分成SNP组和磷酸盐缓冲液(PBS)组,SNP组用不同浓度(0.2、0.4、0.6、0.8、1.0 mg/mL)的一氧化氮(NO)供体药SNP稀释液0.50 mL进行腹腔注射,PBS组使用相同体积的PBS进行腹腔注射,用药15 min后检测外周血中NO及一氧化氮合酶(NOS)的表达,及ITP小鼠外周血ROS浓度。结果 SNP组中,SNP浓度分别为0.2、0.4、0.6、0.8、1.0 mg/mL注射15 min后小鼠血清中NO及NOS的浓度均高于PBS组(P<0.05),NO含量及NOS活性随着用药浓度的升高而升高,差异均有统计学意义(P<0.05),ROS浓度随着SNP浓度的增加而增加(P<0.05),NO表达量与ROS浓度呈正相关(r=0.81,P<0.05),NOS表达量与ROS浓度也呈正相关(r=0.84,P<0.05)。结论在ITP小鼠模型内,NO供体药物SNP以浓度依赖的形式增加ROS的生成,ROS在ITP病理发展中可能与NO具有协同作用,SNP可能通过ROS途径来发挥相应的生理功能。Objective To investigate the impact and significance of different concentrations of sodium nitroprus- side (SNP) on reactive oxygen species (ROS) in idiopathic thrombocytopcnic purpura (ITP) mice. Methods ITP mice were divided into the sodium nitroprusside (SNP) groups, in which different SNP concentrations (0. 2, 0.4, 0. 6, 0. 8 and 1.0 mg/mL) were given intraperitoneally, and phosphate buffered saline (PBS) control group. The NO and NOS in peripheral blood were assessed, while ROS concentrations were also assessed. Results The serum levels of NO and NOS in SNP groups were significantly higher than those in PBS control group ( P 〈 0. 05 ) , so were the NO content and NOS ac- tivity (P 〈0. 05). The NO content and NOS activity were significantly increased as SNP elevated (P 〈0. 05), so was ROS concentration ( P 〈 0. 05 ). The NO and NOS expression were significantly positively correlated with ROS concentra- tion ( r = 0. 81 and 0. 84, respectively, P 〈 0. 05 ). Conclusion NO donor drug SNP promotes the generation of ROS in a concentration - dependent manner in ITP mice. ROS may have a synergistic effect with NO in the pathogenesis of ITP, via which SNP effects.
