摘  要:目的探讨无水化处理在病理组织HE染色中的应用价值,以寻求提高HE染色质量的方法。方法手术切除的人体组织制成蜡块后,切成4μm厚的切片,切片经脱蜡、水化后分成对照组和实验组。对照组行常规HE染色,实验组在切片进入染液前先行无水化处理,再行HE染色。光学显微镜下观察比较染色效果。结果对照组HE染色的切片,细胞核和细胞质染色较淡,细胞核呈浅蓝色,细胞质淡红色,核质对比不清晰,并有少量苏木精沉渣附着;实验组经无水化处理后HE染色的切片,细胞核和细胞质染色较好,细胞核呈深蓝色,细胞质鲜红色,核质对比清晰,无苏木精沉渣附着。结论无水化处理在病理组织HE染色中,可明显提高HE染色质量。Objective To investigate the application value of anhydrous treatment in HE staining of pathological tissue samples in order to improve the quality of HE staining. Methods The human organ samples obtained by exairesis were made into wax blocks and sliced 4 Ixm-thick pieces. After dewaxing and hydration, the slices were divided into control and experimental groups. The slices of the control group were stained with routine HE method and the slices of the experimental group were pre-treated by anhydrous step before HE staining. The staining effects were observed under light microscope and compared between the two groups. Results The dyeing of the karyon and cytoplasm in the slices of the control group was light. The karyon was light blue, and the cytoplasm was light red. The contrast between karyon and cytoplasm was fuzzy, and there were few haematoxylin residues clung to the slices. As to the slices in the experimental group, the dyeing of the karyon and cytoplasm was better. The karyon was dark blue, and the cytoplasm was bright red. The contrast between karyon and cytoplasm was clear, and there was no haematoxylin residues clung to the slices. Conclusion The application of anhydrous treatment to the HE staining of pathological tissue samples can significantly improve the staining quality.
