高效液相色谱法测定止痢宁片中木香烃内酯和去氢木香内酯含量

摘　　要：目的建立止痢宁片木香烃内酯和去氢木香内酯含量测定的高效液相色谱方法。方法色谱柱为AgilentEclipseXDBC18柱（150mm×4．6mm，5μm），以甲醇-水（75：25）为流动相，流速为1．0mL／min，检测波长为225nm，柱温为30℃。结果木香烃内酯进样量在0．2—10mg范围内与峰面积线性关系良好（r=0．9998），加样回收率为99．33％，RSD为0．62％（n=6）；去氢木香烃内酯进样量在0．2—10mg范围内与峰面积线性关系良好（r=0．9999），加样回收率为99．22％，RSD为0．81％（n=6）。结论该方法操作简单，木香烃内酯和去氢木香内酯均能达到较好的分离效果，结果准确可靠，专属性强，重现性好，可用于止痢宁片的质量控制。Objective To establish an high performace liquid chromatography（HPLC） method for the content determination of costunolide and dehydrocostus lactone in Zhilining Tablet, Methods The Agilent Eclipse XDB C18 column（150 mm ×4. 6 mm, 5 μm） was used with methanol-water（75：25） as the mobile phase;the flow rate was 1.0 mL/min; the detection wavelength was set at 225 nm;the column temperature was 30 ℃. Results The sample size of costunolide in the range of 0. 2- 10 mg showed the good linear relation with the peak area （r=0.999 8）,the average recovery rate was 99.33%, RSD 0.62% （n=6）;The sample size of dehydrocostus lactone in the range of 0. 2- 10 mg showed the good relation with the peak area（r=0. 9999）. The average recovery rate was 99.22%, RSD=0. 81% （n = 6）. Conclusion This method is simple to operate, with good separation effect of costunolide and dehydrocostus lactone, accurate and reliable detection results, strong specificity and good reproducibility, which can be used to control the quality of Zhilining Tablet.
