摘 要:目的:建立中药材标准物质DNA图谱,为该药材鉴定提供参考和实验依据。方法:选取25种随机引物,利用随机扩增多态性DNA技术(Random amplified polymorphic DNA,RAPD)对提取出的中药材DNA进行PCR扩增,得到的产物进行琼脂糖凝胶电泳,并对特异性条带进行统计分析。结果:对68种中药材标准物质进行了考察,对其中8种常用于心血管保护的中药材进行了RAPD法DNA图谱的建立,寻找出它们的特异扩增产物。结论:RAPD法可以用来对中药材进行生物学鉴定,所建立的DNA图谱可用来为之后该种药材的鉴定提供依据。Objective : To establish DNA graphs for Chinese medical herbs for providing criterion and refer- ence to identify specific Chinese medical materials. Methods: DNA was extracted from crude medicines, and then random amplification polymorphic DNA (RAPD) method was used to separately amplify genome DNA by 25 single arbitrary primers. The polymorphism PCR products were useful as genetic markers that were separated by agarose gel electrophoresis and detected by staining in gel-stain. Quantity One software was used to analyze the results. Re- sults: This experiment was conducted on 68 standard Chinese medical herbs. Among them DNA polymorphism graphs were set up for 8 medical herbs, and their specific RAPD PCR products were found. Conclusion: RAPD can be used as an effective method to identify Chinese medical herbs. The DNA graphs can be used as reference for these herbs in the future.
