摘  要:目的:观察三草降压汤(SCD)不同组分对H2O2诱导的人脐静脉内皮细胞(HUVEC)氧化损伤模型的形态和活性影响,研究SCD不同组分对HUVEC氧化应激损伤的保护作用,探讨其降压作用的有效组分。方法:采用大孔树脂富集纯化技术,制备SCD的4种组分(分别为SCD-DW、SCD-10ET、SCD-50ET、SCD-95ET),用血清药理学方法分别制备不同组分的含药血清,以5%、10%、15%的含药血清浓度作用于H2O2损伤的HUVEC,于倒置显微镜下观察细胞形态,用四甲基偶氮唑盐法检测细胞活力。结果:SCD各组分均能提高氧化损伤HUVEC的细胞活性,其中SCD-95ET较正常对照组有显著性差异(P<0.05)。结论:SCD的4种组分对H2O2损伤HUVEC均具有不同程度的保护作用,其中SCD-95ET可以作为SCD抗氧化损伤的有效组分进一步深入研究。Objective: To investigate effects of drug serum of Sancao Jiangya Decoction (SCD) different components on morphology and viability of H202-induced oxidative damage model in human umbilical vein endothelial cell (HUVEC), and to study the protective effect of SCD different components on HUVEC oxidative stress injury, as well as to explore an effective component for decreasing blood pressure. Methods: With the enrichment and purification technology of macroporous resin, four groups of SCD components (SCD-DW, SCD-10ET, SCD-50ET and SCD-95ET) were prepared respectively, and then the drugserum of four components was obtained respectively by the serum pharmacological method. HUVEC induced by H202 was treated with 5%, 10%, and 15% of the drug serum concentrations, the cell morphology was observed with inverted microscopy, and the cell viability was examined by methyl thiazolyl tetrazolium assay. Results: Each group of SCD components could enhance the cell viability of HUVEC, of which, the group of SCD-95ET was more obviously than the other groups (P〈0.05). Conclusion: Four kinds of SCD components have varying degrees of protective effects on the H202-induced HUVEC, and SCD-95ET could be used for further study as an effective component of SCD on the function of anti-oxidative damage.
