摘  要:目的:观察电针对坐骨神经损伤大鼠背根神经节中生长相关蛋白-43(GAP-43)的影响,探讨电针治疗坐骨神经损伤的生物学机制。方法:采用大鼠坐骨神经夹持损伤模型,通过光热耐痛阈观察大鼠的行为学变化;通过免疫组织化学染色观察大鼠背根神经节中GAP-43表达情况,进而统计分析各组间的差异。结果:造模后7d模型组大鼠行为学检测表明,坐骨神经损伤后大鼠的感觉功能明显降低(P<0.01);电针治疗10次、20次后,与模型组相比电针治疗组光热耐痛阈(P<0.05)和痛敏分数(P<0.01)明显降低,感觉功能显著恢复。造模后7d模型组GAP-43表达显著升高(P<0.05),电针治疗10次、20后,电针治疗组的GAP-43表达升高,与模型组比较均有显著性差异(P<0.01),与模型对照组相比也有明显升高(P<0.05)。结论:电针可以通过提高相应节段背根神经节中GAP-43的释放,从而促进神经元的发育和再生,最终改善坐骨神经损伤大鼠的感觉功能。Objective: To explore the biological mechanisms of electric acupuncture on the treatment of sciatic nerve injury through the research on changes of GAP-43 expression in DRG of sciatic nerve injury rats. Methods: After making the model of sciatic nerve clamping damage, the variation of rats' behavior were observed through solar-thermal pain threshold resistance. Changes of GAP-43 expression in DRG were observed through immunohistochemical staining, and then statistical analyzed the differences between the groups. Results: Seven days after making the model, compared with normal group, the variation of rats' behavior showed that the sensory function in model group was decreased obviously (P〈0.01). After ten and twenty times of electric acupuncture therapy, both solar-thermal pain threshold resistance and pain sensitivity scores of the therapy group were obviously lower than that of model group (P〈0.05, P〈0.01), and the sensory function was recovered significantly. After seven days of sciatic nerve injured, GAP-43 expression of model group was increased (P〈0.05). After ten and twenty times of electric acupuncture therapy, expression of GAP-43 in DRG from electric acupuncture therapy group significantly increased, staining extent and scope significantly better than model group (P〈0.01) and model control group (P〈0.05). Conclusion: Electric acupuncture could promote the expression of GAP-43 in DRG, the development and the regeneration of injured nerves, in the end enhance the sensory function of the sciatic nerve injury rats.
