摘 要:目的:观察扶正化瘀方对单侧输尿管结扎所致大鼠肾间质纤维化的疗效。方法:70只SD大鼠随机分为正常对照组、假手术组、模型组、氯沙坦钾组及扶正化瘀方(FZHY)低、中、高剂量组,以单侧输尿管结扎术建立单侧输尿管梗阻(UUO)肾间质纤维化模型。观察大鼠一般情况、肾体比;检测肾功能(SCr和BUN),盐酸水解法检测肾组织羟脯氨酸(Hyp)含量;HE染色、天狼猩红染色观察肾组织病理形态。结果:与模型组相比,各治疗组大鼠肾体比不同程度降低(P<0.01),肾组织Hyp含量下降(P<0.01),SCr和BUN含量下降(P<0.01,P<0.05),肾脏纤维化程度明显减轻,其中FZHY中、高剂量药效明显。FEHY中、高剂量组大鼠肾体比、Hyp含量、SCr含量指标变化均优于氯沙坦甲组(P<0.05,P<0.01)。结论:扶正化瘀方能有效抑制UUO大鼠肾间质纤维化,改善大鼠肾功能,且具有一定的剂量依赖性。Objective: To observe the efficacy of Fuzheng Huayu Recipe (FZHY) on renal interstitial fibrosis rats caused by unilateral ureteral ligation. Methods: 70 SD rats were randomly divided into a normal control group, a sham operation group, a model group, a losartan potassium group (LP group) and a low-, medium-, high-dose of FZHY group (LF, MF, HF group), and the renal interstitial fibrosis of unilateral ureteral occlusion (UUO) model was established by unilateral ureteral ligation. The general condition and kidney weight/body weight (KW/BW) of rats were observed; the renal function was detected, including serum creatinine (SCr) and blood urea nitrogen (BUN), and the content of hydroxyproline (HYP) in renal tissues was detected by hydrochloric acid hydrolysis method; the pathological features of renal tissues were examined by hematoxylin-eosin (HE) staining and sirius red staining. Results: Compared to the model group, KW/BW was lowered to varying degrees in each treated group, the content of Hyp in renal tissues, SCr and BUN was decreased, and the degree of renal fibrosis apparently reduced, which was significantly in the MF and HF groups. The KW/BW, contents of Hyp and SCr in the MF and HF groups superior to that in the LP group (P〈0.05). Conclusion: Fuzheng Huayu Recipe has an effective inhibition on renal interstitial fibrosis in UUO rats, and it could improve the renal function with certain dose-dependent manner.
