摘 要:目的:研究榅桲叶提取物对肾性高血压大鼠(RHR)的降压作用以及对血管紧张素Ⅱ(AngⅡ)、肾素活性(RA)、apelin-12、内皮素(ET)、一氧化氮(NO)的影响。方法:采用两肾一夹法(2K1C)制造肾性高血压大鼠(RHR)模型,随机分为6组:假手术组、模型组、榅桲叶提取物高、中、低剂量组、阳性对照药卡托普利组,每组10只。连续给药8周,在给药前和给药后每2周测量大鼠的收缩压(SBP)和舒张压(DBP),末次给药后测量大鼠AngⅡ、RA、apelin-12、ET、NO的变化。结果:给药8周后,榅桲叶提取物明显降低RHR的血压(P<0.05);榅桲叶提取物可以减少血和肾脏AngⅡ和apelin-12的含量,降低RA,减少血浆ET并提高血清NO含量(P<0.05)。结论:榅桲叶提取物的降压作用可能与影响肾素-血管紧张素系统的活性,减少缩血管物质的同时增加舒血管物质的释放有关。Objective: To study the anfihypertensive effect and related substances Cydonia oblonga Mill. (COM) leaf extracts, such as angiotensin-Ⅱ (Ang Ⅱ ), renin activity (RA), apelin-12, endothelin (ET) and nitric oxide (NO). Methods: Twokidney, one-clip (2K1C) model rats were operated and divided randomly into six groups: sham group, model group, captopril group, COM leaf extracts low, medium and high dose groups, 10 rats in each group. The drugs were administered for eight weeks. Systolic blood pressure (SBP) and diastolic blood pressure (DBP) were measured before and every 2 weeks after administration. After the last administration, all rats were sacrificed under deep anesthesia, and blood and kidney samples were collected to detect the level of Ang II,, RA, apelin-12, ET and NO. Results: After treatment for eight weeks, COM leaf extracts significantly declined the pressure of RHR (P〈0.05), which had no significant difference in COM high dose group compared with captopril group; In COM leaf extracts groups, the levels of Ang Ⅱ, apelin-12, RA and ET were declined, while the level of NO was increased. Conclusion: The antihypertensive activity of COM leaf extracts may be related to action on the renin-angiotensin system and balance of the vaso-excitor materials and vasodilator substances.
