摘  要:目的:探讨黄芪对2,4,6-三硝基苯磺酸(TNBS)诱导的大鼠结肠炎的作用。方法:将40只雄性SD大鼠随机分为4组:正常组、模型组、黄芪组、巴柳氮组。除正常组外,其余3组大鼠均以TNBS灌肠造模。灌肠24h后,黄芪组开始每天给予黄芪4g/kg、巴柳氮组给予巴柳氮钠1g/kg灌胃治疗10d。每天观察大鼠一般情况,给药结束后,观察大鼠结肠大体损伤及病理,ELISA法检测肠组织肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)及IL-10水平。结果:两治疗组体质量较模型组增加,但差异无统计学意义;两治疗组DAI评分较模型组明显下降(P<0.05)。黄芪组、巴柳氮组结肠大体形态损伤及病理评分较模型组明显下降(P<0.01)。黄芪组较模型组TNF-α、IL-1β水平明显降低(P<0.05,P<0.01),IL-10水平差异无统计学意义。巴柳氮组较模型组TNF-α、IL-1β、IL-10水平均明显降低(P<0.01)。结论:黄芪能有效改善TNBS诱导的大鼠结肠炎,其机制可能与调节细胞因子水平有关。Objective: To investigate the effects of Radix Astragali on 2,4,6-trinitrobenzenesulphonic acid (TNBS)- induced colitis in rats. Methods: 40 male sprague dawley (SD) rats were randomly divided into four groups: the control group, model group, Radix Astragali group, Balsalazide group. Experimental colitis was induced in rats by enema administration of TNBS except the normal group. Rats in Radix Astragali and Balsalazide groups were treated by gavage with Radix Astragali (4g/kg) or Balsalazide (lg/kg) for ten consecutive days after 24 hours of TNBS administration. General Situation, Macroscopic lesion and histological damage were determined, and cytokine level including interleukin-113 (IL-113), tumor necrosis factor-or (TNF-ct) and 1L-10 were determined by enzyme-linked immunosorbant assay (ELISA). Results: Compared with model group, the rats weight of both treatment groups improved, but there was no statistical difference. The rating of disease activity index (DAI) of both treatment groups were lower than the model group (P〈0.05). The colonic macroscopic morphous and pathohistology of rats in the Radix Astragali group and Balsalazide group improved (P〈0.01). The TNF-ct, IL-1β levels of Radix Astragali group was lower than the model group (P〈0.05, P〈0.01). There was no statistical difference in IL-10 level. The level of TNF-α, IL- 1β and IL-10 in the Balsalazide group were lower than the model group (P〈0.01). Conclusion: Radix Astragali could ameliorate effectively colitis in rats indlBDed by TNBS, and the underlying mechanism may be related to the level of cytokines.
