摘  要:目的:观察心衰合剂对小鼠体外心肌细胞肥大模型细胞凋亡及乳酸脱氢酶(LDH)释放率的影响。方法:培养大鼠乳鼠原代心肌细胞,通过AngⅡ建立肥大心肌细胞模型。Hoechst33342染色检测心肌细胞的早期凋亡率;乳酸脱氢酶漏出率评价细胞受损程度。结果:模型组心肌细胞凋亡率显著提高(P<0.01),LDH漏出率显著上升(P<0.05);5%含药血清组心肌细胞早期凋亡率显著降低(P<0.01);LDH漏出率无明显降低;与5%含药血清组比较,10%含药血清组心肌细胞早期凋亡率显著降低(P<0.01);LDH漏出率降低(P<0.05),结论:与5%含药血清组细胞相比,10%含药血清组细胞更能降低凋亡率及LDH漏出,提示心衰合剂具有最佳剂量,降低药物浓度将会影响其保护作用。Objective: To observe the effects of Xinshuai mixture on vitro hypertrophic myocardial cell apoptosis and LDH leakage rate. Methods: Cultured primary neonatal rat myocardial cells, established hypertrophic myocardial model by AngII. Compared myocytes early apoptosis rate by Hoeehst33342 staining method. Evaluated the degrees of myocardial cell damages by testing LDH leakage rate. Results: Early apoptosis rate of myocardial cells increased significantly (P〈0.01) as well as the release rate of LDH (P〈0.05)in hypertrophic myocardial cell. The early apoptosis rate of 5% Xinshuai mixture group has decline (P〈0.01), but activity of LDH of 5% Xinshuai mixture didn't change significantly. The early apoptosis rate of 10% Xinshuai mixture group was lower markedly (P〈0.01), and release rate of LDH declinedthan 5% Xinshuai mixture group (P〈0.05). Conclusion: Comparing with the 5% Xinshuai mixture group, 10% Xinshuai mixture group can reduce the rate of early apoptotic and release rate of LDH, prompting that there may be the best dose for Xinshuai mixture in clinic, if reducing thedensity of the medicine, efficacy could be worse.
