摘 要:目的:探讨安肺益肾方对熏香烟加气管注内毒素法制造的慢性阻塞性肺疾病(COPD)模型大鼠肺组织匀浆炎性因子白细胞介素--6(IL-6)、IL-8、肿瘤坏死因子--α(TNF-α)的作用,以及抗氧化指标超氧化物歧化酶(SOD)、丙二醛(MDA)的作用。方法:熏香烟加气管注内毒素法建立COPD模型,实验设正常组,模型组,安肺益肾方大、中、小剂量组,氨茶碱组。连续给药4周后,取大鼠左上肺制成10%匀浆液,用Elisa法检测IL-6、IL-8、TNF-α、SOD、MDA含量。结果:与正常组比较,模型组肺组织匀浆中IL-6、IL-8、TNF-α、MDA含量增加,SOD含量降低(P<0.001),安肺益肾方能降低模型大鼠肺组织匀浆中IL-6、IL-8、TNF-α、MDA含量(P<0.05,P<0.01,P<0.001),增加SOD含量(P<0.05)。结论:安肺益肾方对COPD模型大鼠有一定抗炎、抗氧化的作用。Objective: To investigate the effects of Anfei Yishen Formula (AFYSF) on IL-6, IL-8, TNF-a, SOD, MDA of rats pulmonary tissue homogenate with chronic obstructive pulmonary disease (COPD) induced by intratracheal instillation of lipopolysaccharide and passive smoking. Methods: The rat models with COPD were induced by intratracheal instillation of lipopolysaccharide and passive smoking, except normal control group. 72 Wistar rats were divided into AFYSF high, middle, small dose group, aminophylline group, model group, and normal group by a randomized single blind method. The left upper lung of rat was made into 10% homogenate, then the IL-6, IL-8, TNF-a, SOD, MDA levels in the 10% homogenate were tested by Elisa after four weeks' treatment. Results: Compared with the normal group, the IL-6, IL8 TNF-a, MDA levels in the model group were increased (P〈0.001), the SOD level was decreased (P〈0.001). AFYSF can reduce the expression of inflammatory factors in rats pulmonary tissue homogenate with COPD induced by intratracheal instillation of lipopolysaccharide and passive smoking (P〈0.05, P〈0.01, P〈0.001); AFYSF can increase SOD level and decrease MDA level in rats pulmonary tissue homogenate (P〈0.05). Conclusion: AFYSF had anti-inflammatory and antioxidant effects in rats with COPD.
