蕲蛇Ⅱ型胶原蛋白对CIA大鼠的治疗作用

摘　　要：目的:研究蕲蛇Ⅱ型胶原蛋白(CⅡ)对胶原诱导性关节炎(collagen induced arthritis,CIA)模型大鼠的治疗作用,观察其治疗效果并探讨其作用机制。方法:90只Wistar大鼠除空白组外均采用牛CⅡ建立CIA模型并进行随机性分组,分为模型组、牛CⅡ组、蕲蛇水提液高、中、低剂量组、蕲蛇CⅡ高、中、低剂量组,每组10只。造模后第11 d开始给药,期间采用容积法测量关节肿胀程度,连续给药20 d后观察大鼠踝关节病理改变并采用流式细胞术检测大鼠外周血中CD4+T细胞、CD8+T细胞亚群水平。结果:与模型组相比,蕲蛇高、中剂量组、蕲蛇CⅡ高、中剂量组和牛CⅡ组均能显著降低CIA大鼠右后足跖肿胀程度、CD4+T细胞水平,改善大鼠踝关节病理形态,其中蕲蛇CⅡ中剂量组和牛CⅡ组还能显著降低CD4+/CD8+比值(P<0.05)。结论:蕲蛇水提液及蕲蛇CⅡ提取物对CIA大鼠关节炎症均有一定的治疗作用,其治疗机理可能是通过主动免疫抑制降低机体亢进的免疫应答实现的。Objective： To explore the therapeutic effects and mechanisms of type II collagen of Agkistrodon acutus on collageninduced arthritis （CIA） in rats. Methods： The Wistar rats were modeled by bovine type II collagen （C Ⅱ）, then divided into normal g^up, model group, bovine C II group, the aqueous extract and C Ⅱ of Agkistrodon acutus groups with high, middle and low dosages respectively. The paw volume was measured to determine joint swelling degree of the CIA rats. After the model rats treated for 20 days, the blood of all rats were collected to measure the level of CD4 ＊ T ceils, CDs ~ T cells by flow cytometry. The pathological changes of ankle tissue were observed under microscope. Results ： Com- pared with model group, the paw swelling degree and the level of CD4 ~ T cells, CDs ~ T cells were significantly decreased in bovine C II group, high - dose aqueous extract of Agkistrodon acutus, middle - dose aqueous extract of Agkistrodon acutus, high - dose C II of Agkistrodon acutus group and middle - dose C II of Agkistrodon acutus group （ P 〈 0. 05 ）, in these groups, pathological morphology of the rat ankle joint was also improved. Bovine C Ⅱ group and C II of Agkistrodon acutus medial - dose group could significantly reduce the ratio of CD4^＋ / CD8^＋ （ P 〈 0. 05 ）. Conclusion： The aqueous extract and C Ⅱ of Agkistrodon acutus could inhibit the inflammation of CIA rats. The mechanisms that the aqueous extract and type Ⅱ collagen of Agkistrodon acutus could treat rats＇rheumatoid arthritis which may be through active immunosttppression to achieve the purpose of suppressing autoimmune reaction.
