摘  要:目的:通过建立房颤大鼠模型,研究房颤状态下Cx43磷酸化水平对房颤发生的影响以及中药青蒿提取物对Total-Cx43、NP-Cx43表达与分布的影响。方法:建立房颤大鼠模型,使用青蒿提取物进行干预,以卡托普利作为对照组。实验当日处死动物后Western-blot法检测Cx43表达及磷酸化水平。激光共聚焦显微镜检测Total-Cx43、NP-Cx43分布情况。结果:Total-Cx43的表达中对照组的表达量最高,模型组表达量最低,较对照组下降了(57.2±12.5)%(P<0.05),青蒿组与卡托普利组相比,表达量没有统计学差异(P>0.05)。NPCx43的表达中对照组的表达量最高,模型组表达量最低,模型组、青蒿组、卡托普利组之间没有统计学差异(P>0.05)。结论:房颤的发生与Total-Cx43表达减少以及重新分布有密切关系。青蒿提取物以及卡托普利对房颤的改善作用机制之一是通过改变Total-Cx43的表达与分布实现的。在房颤的发生过程中,Total-Cx43的表达与分布发生改变,NP-Cx43的表达与分布无异常,说明P-Cx43是诱发房颤的重要因素。Objective: Through the establishment of rats model of atrial fibrillation, we studied the impact of Cx43 phosphorylalion levels of atrial fibrillation on atrial fibrillation state and the effect of Artemisia annua extract on Total - Cx43, NP - Cx43 expressions and distributions. Method : The rat models of atrial fibrillation were established, using Arte- misia annua extract to intervene and the captopril was as the control group. Experimental animals were sacrificed at the day of experiment and Western - blot was used to assay Cx43 expression and phosphorylation levels. The laser scanning confocal microscope was used to test Total - Cx43 and NP - Cx43 distributions. Result: The highest expression level of Total - Cx43 expression was in the control group, the model group was the lowest. Model group decreased ( 57.2 ± 12.5 ) % compared with the control group( P 〈 0. 05). The expression level in Artemisia annua group was not statistically significant compared with the captopril group ( P 〉 0.05). Highest expression level of NP - Cx43 expression was in the control group, the model group was the lowest. There was no significant difference ( P 〉 0. 05 ) among the model group, Artemisia annua group and captopril group( P 〉 0.05). Conclusion: The incidence of atrial fibrillation is closely related with the reduce and re - distribution of Total - Cx43 expression. By changing the Total - Cx43 expression and distribution, Artemisia annua extract and captopril can improve atrial fibrillation. Total - of Cx43 expression and distribution has changed in the atrial fibrillation process, NP - of Cx43 has no abnormal expressions or distributions, indicating P - Cx43 is an important factor for atrial fibrillation.
