摘 要:目的探讨罗勒多糖抗肿瘤转移作用的可能机制。方法通过Metrigel穿膜法体外观察罗勒多糖对肿瘤细胞迁移的影响;建立不同途径的肿瘤体内转移模型,观察罗勒多糖对肿瘤转移行为的影响;Lucifer Yellow划痕标记荧光传输法检测罗勒多糖对细胞通讯的影响。结果在体外经不同浓度的罗勒多糖处理48 h后的PG细胞,穿过人工基底膜的细胞数显著减少;罗勒多糖对黑色素瘤B16-F10人工肺自发性转移行为具有明显的抑制作用;罗勒多糖对细胞通讯功能缺陷的PG细胞具有一定程度的恢复作用。结论罗勒多糖在体内外均有显著的抗肿瘤侵袭、转移作用,对细胞通讯功能具有一定程度的恢复作用,其机制是通过多个药效靶点综合作用的结果。Objective To explore the possible mechanisms of basil polysaccharide in inhibiting tumor metastasis. Methods Assayed the metastasis of tumor cell ability by Metrigel in vitro. Estabhshed different pathways model of tumor metastasis in vivo, and observed the influence of basil polysaccharide on tumor metastasis behavior. Tested the effect of basil polysaccharide on the recovering of cell communication by Lucifer Yellow scrape loading and dye transfer (SLDT) technique. Results The number of PG cells that went through artificial basement membrane were significantly reduced after treatment with different conce^tra- tions of basil polysaccharide for 48h. Basil polysaccharide had significantly inhibiting effect on melanoma B16-F10 artificial lung spontaneous metastatic behavior; and played a certain role in cell communication recovery. Conclusion The inhibition of tumor cell migration and recovery of tumor cell communication function may be involved in the anti-tumor metastasis effect of Basil polysaccharide.
