摘 要:高效液相色谱法构建复杂中成药黄连上清丸的HPLC指纹图谱。方法首先对实验的方法学进行评价,证明方法精密可靠;用此方法构建了12个样品的图像式指纹图谱;进而构建数字化指纹图,并改进了原有数字化指纹谱的表现形式,使样品间的量化比较一目了然。结论:从图象式指纹谱上可以直观地判断不同样品共有峰的含量差异比较显著,这是由于原料、加工过程等多种因素引起。在数字化指纹谱中,通过对各种参数进行评价,证明每个厂家都按规定投入了十七味原药材,而药材的品质以及药材的投入量却有显著差异。HPLC法构建的黄连上清丸指纹图谱结果有效可靠,该方法可用于其他不同厂家不同批次的制剂控制与评价参考。Objective To develop an effective analysis method by HPLC to analyze a complicated herbal preparation coptis bolus for clearing the upper heat. Methods The methodology validation of HPLC analysis showed the method was accurate and the results were reliable. The method was applied to 12 bolus samples. Image chromatographic fingerprint (ICF) was established first and digitized chromatographic fingerprint spectrum (DCFS) was developed based on the common peaks that selected from the peaks. Conclusion: ICF and DCFS provided fast and practical method for qualitative and quantitative quality control of coptis bolus for clearing the upper heat. The whole concentration distribution of components was shown obviously by ICE The result indicated the instability of quality of the herbal preparation which was related to different producing area, growing condition, climate, harvest time and drug processing and so on. It was possible to extend this methodological approach to quality control of complicated TCM and herbal preparations.
