摘 要:目的:探讨采用全自动组织芯片仪进行组织芯片制作,以及组织芯片在肝癌研究中的应用体会。方法:利用组织芯片仪制备肝癌组织芯片16枚,共包含313例肝癌患者肝癌组织。通过免疫组化方法检测了HLA-DR抗体,并比较其在肝癌癌旁及肿瘤内的表达情况。结果:采用全自动组织芯片仪能更加完好地构建肝癌组织芯片。用此组织芯片检测HLA-DR抗原在肝癌中高阳性表达率为51.1%,而在癌旁组织为76.4%,两者具有统计学差异(P<0.000 1)。结论:全自动组织芯片仪制备的组织芯片能简单、快捷检测相关蛋白在肝癌组织中的表达。组织芯片在肝癌研究中具有高效、快速、方便、经济等特点。Objective:To apply automation tissue an'ay building system in tissue microarray (TMA) construction and explore the application value of TMA technology in hepatocellular carcinoma (HCC) research. Methods: TMA blocks which containing 313 biopsies of HCC tissue were prepared using automation tissue array building system. The expression of HLA-DR protein was examined by conven- tional immunohistoehemistry technique. Results: Sixteen TMA blocks were successfully constructed. HLA-DR protein was analyzed in these TMAs and was found highly expressed in peri-tumoral tissues (51.1%) of HCC as compared with intra-tumoral tissues (76. 4% ; P 〈 0. 000 1 ). Conclusion: TMAs can be simply and quickly constructed with automation tissue array building system. TMA techniques are useful in HCC research with the advantages of high-throughput, rapid, simple, economic and equivalent applications.
