摘  要:目的检测胎牛血清白蛋白(Fetal bovine serum albumin,BSA)在脐带源间充质干细胞溶液的含量以代表异源蛋白在脐带源间充质干细胞溶液中的残留情况的适用性,并通过改进培养条件和增加清洗次数以进一步减少BSA等异源蛋白在脐带源间充质干细胞溶液内的残留。方法运用定量ELISA方法检测了脐带源间充质干细胞溶液中BSA的含量,并探讨改进培养方式与增加清洗次数等方法对干细胞溶液中残留的BSA含量的影响。结果对细胞清洗次数的增加能够有效减少BSA等异源蛋白的残留,并且采用无血清培养基进行干细胞培养是减少BSA等异源蛋白在干细胞溶液内残留量的最有效方法。结论通过检测BSA以代表异源蛋白在脐带源间充质干细胞溶液中残留情况具有适用性;并且增加清洗次数能有效地降低BSA的残留量;采用无血清培养基进行干细胞培养是减少BSA等异源蛋白在干细胞溶液内残留量的最有效方法。Objective To explore fetal bovine serum albumin (Fetal bovine serum albumin, BSA) in the umbilical cord derived mesenchymal stem cells to represent the content of heterologous proteins in the umbilical cord-derived mesenchymal stem cells solution residues applicability,and through improved culture conditions and increase the clean- ing frequency to further reduce the BSA and other heterologous proteins in umbilical cord-derived mesenchymal stem cells. Methods Using quantitative ELISA method to detect residues BSA content in umbilical cord-derived mesenchy- mal stem cells solution and to explore the way of cultivating and increase cleaning times for the influence of residual BSA content in stem cell solution. Results Increased cell washing time can effectively reduce the BSA residue heterolo gous protein,and serum free medium for stem cell culture is the most effective method to reduce such as BSA heterolo- gous protein residues in stem cells. Conclusion By detecting the BSA to represent the heterologous protein in residue of umbilical cord mesenchymal stem cells; and increase cleaning frequency can effectively reduce the residues of BSA; with serum free medium stem-cell cultures is one of the most effective method to reduce the BSA heterologous protein in stem cell solution.
