胶原诱导性和佐剂性大鼠关节炎模型的比较

摘　　要：目的:比较胶原诱导性和佐剂性关节炎制备大鼠关节炎模型的效果。方法:雄性SD大鼠30只,随机分成胶原诱导性关节炎组(CIA组)10只,佐剂性关节炎组(AA组)10只,对照组10只。CIA组以等体积牛Ⅱ型胶原(CⅡ)醋酸溶液与弗氏完全佐剂的混合液注射至大鼠的膝关节腔内;AA组以弗氏完全佐剂注射至大鼠的膝关节腔内;对照组以生理盐水注射至大鼠的膝关节腔内。注射后定期用游标卡尺记录各组大鼠的踝关节肿胀程度。1个月后处死大鼠,检测骨密度,拍摄钼靶片。结果:与AA组、对照组比较,CIA组大鼠膝关节肿胀度增加,骨密度下降,钼靶X线骨质吸收更明显,差异具有统计学意义(P<0.05)。结论:CIA模型较AA模型理想,对研究类风湿性关节炎CIA模型更为适合。Objective:To observe the differences between collagen-induced arthritis(CIA) rat models and adjuvant arthritis(AA) rat models.Methods:30 SD rats were randomly divided into CIA group,AA group and control group.Rats in CIA group were received intra-articular injection of bovine type Ⅱ collagen with Freund's complete adjuvant.Rats in AA group were received intra-articular injection of Freund's complete adjuvant alone.Then diameter of ankle joint of rats was termly measured by vernier caliper.Molybdenum target film of ankle joint was photographed,and bone density of ankle joint was detected.Results:Compared with AA group and control group,diameter of ankle joint in CIA group was increased,bone density was decreased(P<0.05);molybdenum target film of ankle joint showed more damage of bone than that in AA group and control group.Conclusion:CIA model is more suitable for the study of rheumatoid arthritis.
