痔血宁合剂对湿热下注证痔病患者MMP-9、VEGF表达的影响

摘　　要：目的:探讨痔血宁合剂对湿热下注证痔病患者痔组织中基质金属蛋白酶-9(MMP-9)、血管内皮生长因子(VEGF)蛋白及其mRNA表达的影响。方法:60例湿热下注证痔病患者随机分为痔血宁组和对照组,每组30例。痔血宁组患者在门诊先服用痔血宁合剂,1周为1个疗程,1个疗程后予以手术治疗;对照组患者不服药直接予以手术治疗。分别采用免疫组化法和免疫荧光定量法检测两组患者的痔组织中MMP-9、VEGF蛋白及其mRNA的表达。结果:与对照组相比,痔血宁组患者痔组织中MMP-9、VEGF蛋白及mRNA的表达均明显降低(P<0.01)。结论:痔血宁合剂改善痔病患者临床症状的机制可能与其降低痔组织中MMP-9、VEGF蛋白含量和表达有关。Objective： To investigate the influence of ＂Zhixuening Mixture＂ on matrix metalloproteinase-9 （MMP-9） and vascular endothelial growth factor （VEGF） protein and mRNA expressions in treating hemorrhoids of downward accumulation of damp-heat syndrome. Methods： Sixty subjects with hemorrhoids of downward accumulation of damp-heat syndrome were randomly divided into two groups： Zhixuening group in which 30 cases were treated with ＂Zhixuening Mixture＂ for one week before operation, and control group in which 30 eases were treated with surgieal operation direetly. The MMP-9 and VEGF protein and mRNA expressions in haemorrhoids tissues were detected by immunohistoehemistry and real-time fluorescent p01ymerase chain reaction （PCR） methods. Results ： Compared with control group, the expressions of MMP- 9 and VEGF protein and mRNA were obviously decreased in Zhixuening group （P 〈 0.01 ）. Conclusion： ＂Zhixuening Mixture＂ can alleviate the symptoms of hemorrhoids, and its mechanisms may have relationship with decreasing the expressions of protein and mRNA of MMP-9 and VEGF in hemorrhoids tissues.
