摘 要:目的采用乙醇注入法制备重组人生长激素(rhGH)脂质体,并建立包封率的测定方法。方法采用乙醇注入法制备rhGH脂质体,建立葡聚糖凝胶柱分离结合HPSEC法测定包封率;以包封率为指标,通过正交设计优化处方和工艺。结果最优处方为磷脂的用量20.8g/L,磷脂与胆固醇质量比为2∶1,磷酸盐缓冲液的浓度为0.02mol/L。按最佳处方及工艺制得的rhGH脂质体包封率为60.50%,平均粒径185.72nm。结论乙醇注入法制备工艺简单,易于掌握,优选得到的脂质体处方和制备工艺稳定可行。Objective To study on optimum formulation and preparative technology of rhGH liposomes by ethanol injection method. Methods RhGH liposomes were prepared by ethanol injection method; sephadex G- 75 column was used to separate free rhGH from liposomes. The concentration of rhGH was determined by HPSEC. Taking the entrapment efficiency as indexes, formulation and preparative technology were op- timized by orthogonal test. Results Optimum formulation was: phospholipids 20.8g/L, phospholipids- cholesterol 2 : 1, concentration of phosphate buffer 0.02mol/L. The entrapment efficiency of rhGH lipo- somes was 60.50%, and average particle size was 185.72 nm. Conclusion Ethanol injection was simple and easy to control for preparation of rhGH lipsomes.
