微小隐孢子虫感染BALB/c小鼠肠道IL-17的变化

摘  要:将24只3~4周龄雌性BALB/c小鼠随机均分为感染组和对照组(n=12),感染组小鼠接种浓度为1×10^6只/L的微小隐孢子虫卵囊液0.2mL/只,对照组接种0.2mL无菌生理盐水。在感染后7d和14d每组分别断颈处死6只小鼠,取空肠和结肠,对肠道组织培养上清中IL—17A及肠道IL-17A和IL17FmRNA表达水平进行检测。结果表明,与空白对照组相比,感染组BALB/c小鼠空肠和结肠中II.17A的表达量显著升高(P〈0.001);感染组小鼠空肠中IL17FtuRNA表达水平也显著上升(P〈0.001);在感染后7d,结肠IL—17F的表达量显著增加(P〈0.001),而感染后14dIL17F的rnRNA表达水平显著下降(P〈0.001)。Twenty-four female BALB/c mice were randomly selected and divided into infected and control group(n= 12). The infected group were dosed orally with 0.2 mL of 1 )〈 10G/L Cryptospo ridum parvum occyst,and control group mice were dosed orally with 0.2 mL isotonic Na chloride. Six mice were sacrificed on day 7 and 14 post infection each group, collected jejunum and colon samples to study the secretion of IL-17A in supernatants of gut culture and the alteration in mR- NA expression level of IL-17A and IL 17F in intestine. The results showed the expression of IL- 17A in jejunum and colon was increase significantly in BALB/c mice infected by Cryptosporidium parvum compared to control(P〈0. 001). The mRNA level of IL-17F was up-regulated significant- ly in jejunum during Cryptosporidium parvum infection,however increased expression of IL-17F was observed in colon at 7 days post infection, and the mRNA level of IL-17F was makedlys de- creased on day 14 post infection in colon.
