血管紧张素转化酶2(ACE2)的基因克隆及在仔猪体内的表达分布

摘　　要：克隆验证了血管紧张素转化酶2(ACE2)基因的核苷酸序列,采用RT-PCR和免疫组化的方法在mRNA和蛋白水平上明确ACE2在仔猪各组织中的表达和分布。RT-PCR显示:ACE2在仔猪体内分布广泛;尤其在肾脏、胃底、回肠、十二指肠、空肠中都有较高水平的表达;免疫组化结果显示:ACE2在胃底部、小肠各部位的肠上皮细胞刷状缘均有显著表达,但在结肠上皮细胞没有ACE2表达。ACE2在胃肠道中有较高水平的表达,提示ACE2可能参与了断奶仔猪胃肠道的生长发育。To investigate the expression of angiotensin-converting enzyme 2 （ACE2） gene in pig lets,especially the distribution and expression in the gastrointestinal tract, the experiment first cloned and verified the nucleotide sequence of the ACE2 gene, then used RT PCR and immunohis tochemical to detecte the expression and distribution of ACE2 in every tissue, mRNA of ACE2 in piglets widely distributed, especially, in kidney, stomach, ileum, duodenum, the middle and posteri or part of jejunum had higher levels of expression, the ACE2 protein distribution in every organs had an apparent specificity. That is, in the gastric gland, brush border of the proximal tubular cells and collecting duct had obvious expression;in small intestine,intestinal epithelial cell brush border of every part had expression,but not ACE2 expression in colonic epithelial cells. ACE2 in the gas- trointestinal tract had higher levels of expression, suggesting that ACE2 may be involved in the growth and development of the gastrointestinal system of weaned piglets.
