摘  要:用枯草芽孢杆菌(Bacillus subtilis)OKB105菌株菌悬液处理水稻种子及幼苗,发现OKB105菌株能够显著促进水稻生长。为了研究枯草芽孢杆菌的促生机制,建立了水稻与OKB105菌株的互作体系,并对互作后的OKB105菌株胞内全蛋白的双向电泳条件进行了优化。结果表明:分离胶浓度为10%,等电聚焦(IEF)程序中最高电压8 000 V,聚焦60 000 Vh且增加低电压慢速除盐步骤能够提高双向电泳图谱质量,为蛋白质组学水平上研究水稻与枯草芽孢杆菌的互作奠定了良好的基础。The rice seeds and seedlings were treated by the suspension of Bacillus subtilis strain OKB105, this study found that the strain OKB105 can significantly promote plant growth. In order to study the growth-promoting mechanism of Bacillus subtilis, this article established interaction system between rice and strain OKB105, optimizing the two-dimensional electrophoresis conditions about total protein of the strain OKB105 after interaction with rice. The results showed that concentration of separating gel 10%, maximum voltage 8 000 V, focus 60 000 Vh, then increasing the step of low voltage and slow desalination could improve the quality of the picture. It will lay a good foundation for studying the interaction between rice and Bacillus subtilis on proteomic level.
