产油脂耐盐真菌Salicorn 5的分离鉴定与发酵产物表征

摘  要:对分离自盐生海芦笋的1株代号为Salicorn 5的耐盐真菌进行鉴定,提取总DNA后分别PCR扩增18S rDNA和ITS1-5.8S-ITS2区域,测序并进行同源性分析,构建ITS基因系统发育树,同时结合形态学分析,将该菌鉴定为小克银汉霉属(Cunninghamella sp.)。该菌经静置发酵后,用环己烷提取发酵产物,并用硅胶柱层析分离得到3个油脂单体化合物,化合物经核磁共振(NMR)和质谱(MS)解析为油酸、亚油酸和1,3油酸双甘油酯。该研究结果说明,小克银汉霉属Salicorn 5菌株能够高产不饱和油脂,具有潜在的工业化应用价值。In this study,a halotolerant fungus Salicorn 5 isolated from the halophyte Salicornia bigelovii was identified. 18S rDNA and ITS1-5.8S-ITS2 region were amplified and sequenced respectively after the extraction of genomic DNA. Homology analysis was conducted and phylogenic tree based on ITS gene sequence was constructed. Combined with morphological analysis, the strain was identified as Cunninghamella species. The fermentation products of this strain were extracted using eyclohexane after fermentation, and three compounds were obtained by silica gel column chromatography. Their structures were elucidated as oleic acid, linoleic acid and 1,3 oleic acid diglyceride on the base of nuclear magnetic resonance (NMR)and mass spectrometry (MS)analysis. The results suggest that Cunninghamella sp. can produce unsaturated oil, and has potential industrial applications.
