摘 要:目的探讨豚鼠耳蜗缺血再灌注损伤后细胞凋亡的情况、凋亡相关蛋白FasL表达的改变以及二者之间的关系。方法健康豚鼠40只随机分成正常组、假手术组和模型组,模型组再分为缺血30min、再灌注6h及24h三组。用组织化学方法观察缺血及再灌注不同时间段耳蜗各部位的组织学改变,用免疫组织化学法检测内耳FasL的表达,用原位凋亡法(TUNEL法)检测内耳细胞凋亡的情况。结果正常组、假手术组内耳罕见凋亡细胞,FasL为弱阳性表达;模型组Corti器、血管纹、螺旋神经节在缺血及再灌注每个时间段均有细胞凋亡,FasL为阳性表达,与正常组比较明显增多(<0.05)。结论 FasL参与了豚鼠耳蜗缺血再灌注损伤后细胞凋亡的发生。Objective To study the expression of apoptosis-related protein Fas ligand (FasL) in cochlea of guinea pigs and its relationship with apoptosis following ischemia-reperfusion injury. Methods Forty guinea pigs were selected and randomly divided into five groups: normal groups, sham control group, ischemia 30 minutes group, ischemia- reperfusion(I/R) 6 hours and 24 hours groups. HE staining, immunohistochemisty and TUNEL were used. Results In the ischemia and I/R groups, the expression levels of FasL in Corti organ, stria vascularis and spiral ganglion were increased significantly compared with the normal and sham control groups, with more apoptosis ceils in the ischemia and I/R groups than in the normal and sham control groups. Conclusion The overexpression of FasL is related to cell apoptosis in the cochlear of guinea pigs after ischemia-reperfusion injury.
