鸡胚分离甲型H1N1流感病毒的特征分析

摘　　要：目的了解鸡胚法分离甲型H1N1流感病毒的特征,为监测实验室开展常规病毒分离和疫苗制备提供参考。方法收集经核酸检测阳性的甲型H1N1流感病例呼吸道标本接种SPF鸡胚,应用红血球凝集试验判定分离结果并测定病毒滴度,以血凝抑制试验鉴定毒株型别。结果对2009-2012年采集的335例甲型H1N1流感病例标本进行鸡胚病毒分离,平均分离率为87.76%。在鸡胚传代第1代,病毒分离阳性率为6.8%,第2代阳性率为64.29%,第3代则达到96.26%。但病毒在鸡胚中的复制效率不高,血凝滴度普遍较低,73.13%的分离物血凝滴度低于1∶16,48.64%的分离物血凝滴度低于1∶4。结论鸡胚对甲型H1N1流感病毒敏感,可用于病毒分离,持续传代有利于提高病毒分离阳性率,但病毒培养物的血凝滴度普遍较低。Objective To study the characteristics of the pandemic influenza A （H1N1） viruses isolated by chicken embryo eggs from 2009 to 2012 in Fujian, China and provide guidelines and references for influenza surveillance and vaccine research. Methods H1N1 virus strains were isolated from chicken embryo-eggs and further characterized. The respiratory specimens were collected from patients confirmed as H1N1. Viral specific nucleic acid was detected and virus was inoculated subsequently into the specific pathogen free （SPF） chicken embryo eggs. The hemagglutination （HA） test was applied for the identification of viral isolates and the titra- tion. Hemagglutination inhibition （HI） assay was used to identify the virus types. Results Virus isolation in chicken embryo eggs was done using H1N1 isolates from 335 patients during the period of 2009 to 2012. The positive rates of viral isolation was 6.8%, 64.29%and 96.26% after the first, second and third passages in SPF chicken embryo-eggs. However, HA test showed that prolonged virus replication was poor in embryo eggs. 73.13% of viral cultures showed the HA titers lower than 1 ： 16, and 48.64% of cultures below 1 ： 4. Conclusions Chicken embryo eggs are sensitive to H1N1 passaging may increase the positive rate of viral isolation; 10W. virus and can be used for virus isolation.
