摘 要:目的 观察正常和胃伤害性刺激大鼠下丘脑乳头体前区 (PMH)内 P物质 (SP) m RNA阳性神经元的分布和变化 .方法 福尔马林导入胃制成内脏伤害性刺激模型 ,用原位杂交组织化学法观察 .结果 对照组的 PMH内 SP m RNA阳性神经元的数量较少 .中等强度的 SP m RNA杂交信号主要见于乳头体内侧核的中间部和外侧部、乳头体外侧核和弓状核 .给予胃伤害性刺激后 ,PMH内表达 SP m RNA的神经元的数量明显增加 ,较强的杂交信号见于乳头体上交叉腹外侧部 ,乳头体内侧核内侧部、中间部和外侧部 ,结节乳头体核尾侧部 ,乳头体前核腹侧部和背侧部 ;中等强度的杂交信号见于弓状核、乳头体上核和乳头体外侧核背侧部 ;较弱的杂交信号见于下丘脑中介核、结节大细胞核 ,下丘脑背内侧核、下丘脑后区及背侧部、丘脑底核、穹窿背外侧部、下丘脑外侧区和乳头体被盖区吻侧部 .结论 大鼠下丘脑乳头体前区内有表达 SP m RNA的神经元 ,其中的部分神经元可能参与了 SP介导的。AIM To observe the distribution and changes of neurons expressing substance P (SP) mRNA in the rat premamillary region of the hypothalamus (PMH) by visceral noxious stimulation to the stomach. METHODS A small amount of formalin was inducted directly into stomach through a plastic tube to make a visceral noxious stimulation model. The results were examined by in situ hybridization histochemical method with SP mRNA probe. RESULTS In normal rats, the number of SP mRNA positive neurons was quite limited. A moderate amount of SP mRNA positive neurons was detected in the median and lateral parts of the medial mamillary nucleus, the lateral mamillary and arcuate nuclei. After noxious stimulation to the stomach, neurons expressing SP mRNA remarkably increased in the PMH. Neurons with heavy SP mRNA signals were noted in the ventrolateral supramamillary decussation, the medial, median, and lateral parts of the medial mamillary nucleus, tuberomamillary nucleus at caudal level, the dorsal and ventral parts of the premamillary nucleus. SP mRNA positive neuronal density was moderate in the arcuate nucleus, supramamillary nucleus, the dorsal part of lateral mamillary nucleus. Scattered SP neurons were also observed in the terete hypothalamic nucleus, tuberal magnocellular nucleus, dorsomedial nucleus, posterior hypothalamus area, subthalamic nucleus, dorsolateral area around fornix, lateral hypothalamus area and mamillotegmental area at rostral level. CONCLUSION There are SP mRNA positive neurons within PMH, some of which might be involved in the regulation and control of the general visceral sensory information transmission.
