摘 要:目的:观察眼镜蛇毒对大鼠脊髓Nestin和BDNF表达的影响。方法:24只大鼠随机分为正常对照组、生理盐水组和眼镜蛇毒组,每组8只。眼镜蛇粗毒以0.5μl/kg注入左后肢内侧复制蛇伤模型,30 min后处死动物,采用免疫组织化学方法,检测脊髓腰段Nestin和BDNF的表达。结果:眼镜蛇毒组大鼠脊髓腰段前角和后角Nestin和BDNF阳性神经元数量较正常对照组、生理盐水组均明显增多(P<0.05),且着色较深,眼镜蛇毒组脊髓Nestin和BDNF平均灰度值较正常对照组、生理盐水组表达均明显降低(P<0.05)。结论:眼镜蛇毒致伤后,脊髓灰质Nestin和BDNF表达显著增加,可能参与脊髓损伤的代偿性修复。Objective:To observe the effects of naja naja atra venom on the expression of nestin and brain-derived Neorotrophin factors(BDNF)in the spinal cord of rats.Metho ds: Twenty-four rats were randomly divided into groups of normal controls,administration of either normal saline or snake venom( n=8 for each) .Poisoning rat models were developed by in-jection of the crude venom of cobra at dosage of 0 .5 μl/kg via left poste-rior limb,and killed in 30 min.Immunohistochemical technique was used to detect the expression of nestin and BDNF in lumbar spinal cord . Results:The number of nestin and BDNF positive cells were increased significantly in the anterior horn and posterior horn of the gray matter in lumbar spinal cord in rats treated with the naja naja atra venom as com-pared with the controls and saline management group(P〈0.05),and the cells presented with darker coloration,whereas the average gray value of nestin and BDNF was notably decreased in the snake venom treated ani-mals than the normal controls and saline groups(P〈0.05).Conclusion:The nestin and BDNF expression were upregulated in the gray matter in spinal cord of rats treated with naja naja atra venom,suggesting that this protein and the factor may be involved in compensatory repair of the in-jured spinal cord.
