摘 要:赖氨酸作为家畜限制性必需氨基酸,其饲料含量直接影响到家畜蛋白质合成。试验利用基因工程方法,从辣椒花药中克隆出高Lys蛋白基因(Cflr),与载体pHT43构建成重组表达质粒,将其转入枯草芽孢杆菌中,IPTG诱导表达,基因在mRNA水平有表达,SDS-PAGE中出现一条明显的蛋白条带,大小为24 ku;经HPLC检测赖氨酸含量,重组菌赖氨酸含量比野生型菌株提高16.05%。该重组菌为以后其作为饲料添加剂进一步应用提供技术基础。Lysine is a restrictive essential amino acid of livestock, its content in feed directly affects livestock protein synthesis. In this research, the lysine- rich protein gene (Cflr) was cloned from pepper anther, and connected with expression vector pHT43, the recombinant plasmid pHT43/Cflr was constructed. The recombinant plasmid was transferred into Bacillus subti/is, upon IPTG induction, mRNA of cflr was observed, and their has an obvious protein band by SDS-PAGE; The content of lysine was detected by HPLC, the content of lysine in recombinant bacterium rised by 16.05% than wild bacterium. This experiment provides research-basis for application of the recombined Bacillus subtilis as feed additive.
