摘 要:目的探讨大鼠颅眶联合伤(COI)后视网膜神经节细胞(retinal ganglion cells,RGC)活性氧含量与凋亡的关系。方法 88只SD大鼠随机分为损伤组(n=64)和无损伤组(n=24)。损伤组大鼠左眼建立COI模型(模型侧),右眼作为自体对照(对照侧),分别在造模后3 d、6 d、10 d、14 d取视网膜组织。检测RGC活性氧含量和不同时间点视网膜细胞凋亡率。光镜下观察视网膜微观结构,电镜下观察RGC超微结构。结果造模后模型侧RGC的活性氧含量和视网膜细胞凋亡率逐渐升高,10 d达到高峰,与对照侧和无损伤组比较,差异有统计学意义(P<0.05)。模型侧视网膜结构紊乱,RGC超微结构表现出典型的凋亡特征。结论 COI后RGC凋亡失调,活性氧含量增高在其中起到重要作用。Objective To investigate the relationship between the content of reactive oxygen species in retinal ganglion cells (RGCs) and cell apoptosis in rats with experimental cranio-orbital injury (COD. MethOds Eighty-eight SD rats were divided into injury group (n = 64) and non-injury group (n = 24). Left eyes of injury group rats were treated as COI model (model side) and right eyes were treated as control side. The retina of rat was obtained at 3, 6, 10 and 14 day respectively after injury. The content of reactive oxygen species and retinal cell apoptosis rate were checked at different time points. The microstructure of RGC was observed under optical microscope. The ultrastructure of RGC was observed under transmission electron microscope. Results The content of reactive oxygen species and the apoptosis rate of retinal cells increased gradually and reached the maximum on 10 day after injury in the model side. There was a statistical difference between model side and control side or non-injury group (P〈 0.05). The retinal structure of model side was disorganized after injury. The ultrastructure of RGC in model side showed the typical characteristics of apoptosis. Conclusions The RGC shows characteristics of apoptosis imbalance after COI and increase of reactive oxygen species content play an important role in apoptosis imbalance of RGCs.
