摘 要:建立了中华鳖日本品系组织中氟甲喹的高效液相色谱-串联质谱检测方法,研究了氟甲喹药饵多次给药后在中华鳖日本品系体内的残留代谢规律.在(25.0±1.5)℃下,采用纯度不小于98%的氟甲喹和规格为(250±50)g的中华鳖日本品系180尾,混饲投喂12mg/kg体重药饵,给药7d.在停药后的第1,2,4,8,16,28,48,72,96,120,144,168,240,360h分别取其血液、肌肉、肝脏和肾脏样品进行残留代谢规律的研究.结果表明氟甲喹在中华鳖日本品系的血液、肝脏、肌肉和肾脏组织中均有残留,最高峰值均出现在停药后2h,质量分数分别为2715.0,2 000.0,1 095.3,281.0μg/kg;其t1/2α分别为1.015,0.629,0.719h,0.212h,t1/2β分别为49.636,52.234,42.817,46.657h,动力学方程分别为:C血液=2 674.136×e-0.683t+1 410.516×e-0.014 0t,C肝胰腺=2429.647×e-1.103t+1 280.742×e-0.013 3t,C肌肉=946.592×e-0.964t+770.039×e-0.016 2t,C肾脏=0.487×e-3.275t+224.740×e-0.014 9t.说明氟甲喹在中华鳖日本品系体内吸收迅速,在各组织中残留量及消除速度差别较大,建议休药期为10d以上.To develop a high performance liquid chromatography-tandem mass spectrometric method for flume- quine residue determination in tissues from Chinese soft-shelled turtle (Pelodiscus sinensis) Japanese strain and to investigate the elimination of flumequine residues in the tissues, 180 healthy soft-shell turtles were administered flumequine at a dosage of 12 mg/kg for 7 d under the temperature of 25.0±1.5 ℃. Samples were slaughtered at different intervals after the last feeding and tissues including blood, muscle, liver and kidney were collected at 1 h, 2 h, 4 h, 8 h, 16 h, 28 h, 48 h, 72 h, 96 h, 120 h, 144 h, 168 h, 240 h and 360 h for pharmacokinet- ics research. The results showed that flumequine residues were found in plasma, liver, muscle and kidney and the maximum content was 2715μg/kg, 2000 μg/kg, 1 095.33 vg/kg and 280.98μg/kg, respectively, 2 h after the last feeding. The absorption half-time (tl/2a) was 1. 015 h, 0. 629 h, 0. 719 h and 0. 212 h, the elimination half life (tl/2β) was 49. 636 h, 52. 234 h, 42. 817 h and 46. 657 h, respectively, and the concentration-time curves of flumequine in blood, liver, muscle and kedney were Cb =2 674. 136 × e ^-0.683t +1 410. 516×e^-0.0140t, C1=2 429. 647×e^-1.103t +1 280. 742×e^-0.0133t, Cm=946. 592×e^-0 .0964t+70. 039× e^- 0.0162t, and Ck=0.487×e^-3.275t+224.740×e^-0.0149t, The results demonstrated that flumequine was ab-sorbed fast in the body of P. sinensis Japanese strain and that the residual quantity and elimination rate of flumequine were significantly different in various tissues. The withdrawal period should not be less than 10 d under the conditions of this experiment.
