蒙药色玛-3汤对大鼠慢性肾衰模型的作用及其对TNF-α的影响

摘　　要：目的:研究蒙药色玛-3汤对大鼠慢性肾衰模型的治疗作用,探讨其作用机制。方法:将50只Wister大鼠随机分为5组:空白对照组、病理模型组、色玛-3汤高剂量治疗组(20g·kg-1·d-1)、色玛-3汤中剂量治疗组(10g·kg-1·d-1)、色玛-3汤低剂量治疗组(5g·kg-1·d-1),每组10只。以腺嘌呤200mg·kg-1·d-1灌胃建立CRF模型,造模同时各治疗组灌胃给予章古-3汤。连续灌胃24d后,检测生化学指标尿素氮(BUN)、血清肌酐(Scr);观察肾组织病理改变;用免疫组化方法检测肾组织中TNF-α的表达和分布。结果:经色玛-3汤治疗的各组大鼠与同期病理模型组比较,肾功能(BUN,Scr清除水平)有明显的改善(P<0.05),病理改变较轻。治疗组TNF-α表达水平显著低于同期病理模型组(P<0.05)。结论:蒙药色玛-3汤大鼠CRF模型的肾功能有一定保护作用,其作用机制可能与抑制TNF-α的表达有关。Objective ：To study the therapeutic effects of Traditional Mongolian Medicine sema - Ill on chronic renal failure （CRF） in rat and explore the mechanism of sema - III in preventing and treating CRF. Methods ： Fifty wister rats were randomly divived into five groups , which including：normal group , model group , sema - III high dosage group, sema - III middle dosage group, sema - III low dosage group, each group had 10 rats. Each rat in model group, sema - III groups were infused with adenine （200oKg - lod - 1 ） into stomach, in the same time ,each rat in sema - III group was also iiffused with high（20g ·kg-1·d-1） ,middle（ 10g ·kg-1·d-1 ）, low（5g ·kg-1·d-1 ） dosage forms sema - III into stomach for 24 days . The content of blood urea nitrogen（BUN） ,serum creatinine （Scr） , and changes in histopathology were observed after 24 days. Tumor Necrosis Factor- α（ TNF-or） expression was examined by immuohistochemistry. Resuhs： in sema - III treated group , the renal function,including the renal clearance level of BUN and Scr is obviously lower than that in model group（ P 〈 0.05）. pathological changes of sema - III treated group were less than the model group. TNF - α expression level is lower compared with model group. Conclusion ： Traditional Mongolian Midicine sema - III can protect renal function of CRF rats. the mechanism may be relative to suppressing the expression of TNF - α.
