摘 要:目的鉴定长春地区孕妇感染弓形虫株的基因型。方法采用间接ELISA法检测长春地区840名孕妇血清中抗弓形虫IgG抗体,阳性者提取全血DNA,以弓形虫529bp高度重复序列为靶基因进行PCR扩增,用多基因位点聚合酶链反应-限制性片段长度多态性(PCR-RFLP)分析鉴定PCR阳性标本的基因型。结果 ELISA检测80份标本IgG阳性,其中33份标本同时PCR阳性。对PCR阳性标本进行PCR-RFLP分型鉴定,有8份标本为基因Ⅱ型。结论长春地区孕妇感染的弓形虫主要为基因Ⅱ型。Objective To identify the different genotypes of Toxoplasma gondii associated with toxoplasmosis in pregnant women in Changchun. Methods Serum samples from 840 pregnant women were examined indirect ELISA for levels of anti-T, gondii IgG. DNA samples extracted from the peripheral blood of IgG-positive pregnant women were used in PCR to detect 529-bp repeat sequences of T. gondii. Genetic characterization of T. gondii isolates was carried out using multilocus polymerase chain reaction-restriction fragment length polymorphisms(PCR-RFLP). Results Anti-T. gondii IgG was detected in serum samples from 80 pregnant women. Thirty-three samples were positive according to PCR. Eight samples were genotyped as Type II. No other genotypes were identified. Conclusion Type II is the main genotype of T. gondii associated with toxoplasmosis in pregnant women in the Changchun area.
