摘  要:目的观察人参皂苷Rb1(Rb1)抗血管平滑肌细胞增殖效应与钙调神经磷酸酶(CaN)信号通路的关系,并探讨该药抗增殖效应是否与其抗氧化作用有关。方法采用球囊导管损伤血管内皮造成大鼠颈动脉内膜增生,ip Rb1两周后,取血样测定过氧化物歧化酶(SOD)活性及丙二醛(MDA)含量变化;在培养的血管平滑肌细胞(VSMC)中加入血小板衍化生长因子-BB(PDGF-BB)诱导其增殖,给予Rb1 24 h后,观察VSMCs增殖和CaN mRNA表达的改变。结果 10、30 mg·kg-1·d-1Rb1给药两周使球囊损伤而变窄的大鼠颈动脉管腔面积明显扩大,伴有SOD活性升高和血中MDA水平的降低;在培养VSMCs中,加入抗增殖浓度Rb1(25、50、100μg·mL-1)后,显著下调PDGF-BB所致CaN mRNA的过度表达。结论 Rb1抗VSMC增殖效应至少部分与其抗氧化作用有关,并可能涉及其对CaN通路的抑制。OBJECTIVE To investigate whether the anti - proliferative effect of ginsenoside Rbl ( Rbl ) on vascular smooth muscle cells (VSMCs) was involved in the inhibition of ealeineurin (CaN) signaling and its anti - oxidant action. METHODS The carotid neointimal hyperplasia model of rat was induced by rubbing the endothelia of vessel with a balloon, and the plasma superoxide dismutase (SOD) activity and malondialdehyde (MDA) eontant were detected after Rbl -administration(ip) two weeks later. The platelet derived growth factor- BB( PDGF -BB) was added in cultural VSMCs to elicit proliferation,and the CaN mRNA expression was exam- ined by real time RT - PCR at 24 h after Rbl - administration. RESULTS Administration of Rbl ( 10,30 mg· kg- l · d - 1 ) for two weeks could widen the narrowed lumen of the rat carotid artery induced by balloon - injury, with increase SOD activity and decrease MDA eontant in the plasma. Rbl at the concentrations that could inhibit the cultural VSMC proliferation and also could down - regulate the CaN mRNA expression by PDGF - BB. CONCLUSION The anti - proliferative effect of Rbl on VSMC is partly related to its anti -oxidant action and may be involved in the inhibition on CaN signaling.
