摘  要:目的 提高毛细管电泳法的灵敏度,建立测定中成药中微量士的宁的高效毛细管电泳法.方法 采用高效毛细管电泳场放大富集法,运行缓冲液为70 mmol· mL-硼砂-乙腈(83:17,pH5.5),10 kV电动进样,进样时间16 s,分离电压25 kV,分离温度25 c℃,检测波长205 nm.结果 士的宁0.2~16.0 μg·mL-1与峰面积的线性关系良好(r=0.9987),检测限为0.022μg·mL-1.结论 所用方法准确、简便,有效提高了毛细管电泳法的灵敏度.OBJECTIVE To improve the sensitivity of capillary electrophoresis and establish an HPCE method for determination of the trace strychnine in Chinese patent medicine. METHODS Field - amplified capillary electrophoresis was used with the running buffer of 70 mmol·mL-1 boric acid- acetonitrile(83 : 17)(pHS. 5) and an applied voltage of 25 kV at 25℃. The detection was performed with diode array detection at 205 nm. The sample was eventually diluted by ultra - pure water and injected for 16 s in electrokinetic type. RESULTS The linearity of strychnine was obtained over the rang of 0.2~16.0 μg·mL-1 ( r = 0. 9987 ) and the detection limit was 0.022μg·mL-1. CONCLUSION The method is simple,sensitive and effectively improved the sensitivity of capillary electroDhoresis.
