摘  要:目的 采用HPLC法测定墨旱莲配方颗粒中原儿茶酸、原儿茶醛的含量.方法 采用正交试验进行提取方法的研究.采用Agilent Zorbax Eclipse XDB-C18柱(150 mm×4.6 mm,5μm),甲醇-0.1%磷酸水溶液(10∶90)为流动相,流速1 mL·min-1,检测波长280 nm,柱温为35℃.结果 采用20倍量75%甲醇,超声提取1h可较好地提取墨旱莲配方颗粒中的原儿茶酸与原儿茶醛.原儿茶酸与原儿茶醛的线性范围分别为33.84 ~112.80(r =0.9999)、3.328 ~16.64μg·mL-1(r=0.9999),平均加样回收率分别为99.50%、100.83%,RSD分别为0.88%、1.53%(n=6).结论 所用方法结果准确可靠、重复性好,为控制墨旱莲配方颗粒的质量提供科学依据.OBJECTIVE To establish an RP - HPLC method for the determination of protocatechuic acid and protocatechualdehyde in dispensing granule of Eclipta prostrata L.. METHODS The extraction method was optimized by orthogonal experiment and the con- tents were determined by HPLC. Eclipse XDB - C18 column(150 mm×4.6 mm,5μm) was adopted. The mobile phase consisted of methanol -0.1% phosphoric acid( 10 : 90) , and the flow rate was 1.0 mL·min-1. The UV detection wavelength was at 280 nm, and column temperature was 35℃. RESULTS 20 times amount of 75% methanol ultrasonic extracting 1 h was the optimal extracting method. The two components had good resolution. The liner ranges were in the range of 33.84 - 112.80 μg·mL-1 ( r = 0. 9999 ) for protoeatechuic acid and in the range of 3. 328 - 16.64 μg·mL-1 ( r = 0.9999 ) for protocatechualdehyde, respectively. The average recoveries were 99.50% and 100.83% with RSD O. 88% and 1.53 % ( n = 6). CONCLUSION The method is accurate,reproducible, and can be used to control the quality of dispensing granule of Eclipta prostrata L..
