摘 要:目的:探讨硫酸软骨素酶ABC(chondroitinase ABC,ChABC)与脂肪源性干细胞(Adipose-derived stem cell,ADSC)联合应用对脱细胞异体神经支架移植修复坐骨神经缺损后大鼠运动和感觉功能恢复的影响。方法:成年大鼠随机分为脱细胞坐骨神经(acellular rat sciatic nerve,ARSN)组、ChABC组、ADSC组、ChABC+ADSC组;应用坐骨神经功能指数(SFI)、电生理和胫前肌湿重比检测神经再生和运动功能的恢复;脊神经节HRP逆行标记及患侧足掌皮肤触觉小体HE染色检测感觉功能的恢复。结果:与ARSN组比较,ChABC组、ADSC组和ChABC+ADSC组SFI显著增高,神经传导速度显著增高,动作电位波幅增大,胫前肌湿重比率增高,HRP阳性标记感觉神经元和触觉小体的数量增高(P<0.05)。其中ChABC+ADSC组作用显著优于单独治疗组(P<0.05)。结论:ChABC联合ADSC移植对大鼠坐骨神经缺损脱细胞异体神经支架修复后运动和感觉功能恢复的作用优于单独治疗组。Objective: To study the effects of chondroitinase ABC (ChABC) combined with adipose-derived stem cell (ADSC) on motor and sensory functional recovery after acellular nerve allograft repair of the sciatic nerve gap in rats. Methods: Adult rats were randomly divided into acellular rat sciatic nerve (ARSN) group, ChABC group, ADSC group and ChABC + ADSC group. The sciatic functions index ( SFI), electrophysiology and the ratio of wet weight of tibialis an- terior muscle were used to evaluate nerve regeneration and motor functional recovery. Horseradish peroxidase (HRP) ret- rograde tracing and hematoxylin-eosin staining (HE) staining of the tactile corpuscles in injured foot skin were used to e- valuate the sensory functional recovery. Results: Compared with ARSN group, SFI, nerve conduction velocity, wave am- plitude, ratio of tibialis anterior wet muscle weight, the number of HRP positive sensory neurons and the tactile corpuscles were significantly increased in ChABC group, ADSC group and ChABC + ADSC group ( P 〈 0.05 ). Moreover, the effects of ChABC + ADSC group were more powerful than those with single drug treatment. Conclusion: The effect of combined ChABC and ADSC transplantation treatment on motor and sensory functional recovery was much better than either of the single treatment after aeellular nerve allograft repair of the sciatic nerve gap in rats.
