摘  要:针对凝胶图像制作过程中会产生蛋白质点重叠和堆聚的问题,提出一种基于形状特征的凝胶图像重叠蛋白质点自动分离算法。首先,采用内外标记分水岭法进行粗检测,提取蛋白质点的初始轮廓;其次,结合极小值深度和形状特征信息自适应设定阈值提取形状标记,并利用提取的形状标记计算形状距离图;然后,采用形态学极小值强加技术,将形状标记强制为形状距离图的极小值;最后,再次采用分水岭法对修改后的形状距离图进行图像分割,实现重叠蛋白质点的精确分离。通过对合成凝胶图像和不同类型的真实凝胶图像进行实验,结果表明,与其它的分水岭改进算法相比,该算法具有较高的重叠蛋白质点正确分离率,其误差小于6%,同时能够获得更为理想的重叠蛋白质点分离效果。In order to detect the overlapped and clustered protein spots in protein gel images, we suggest an auto sep arating algorithm based on the shape characteristics in this paper. Firstly, we used a marker-controlled watershed al gorithm to detect the initial outline of the protein spots in application of the method. Secondly, we extracted shape markers adaptively by combining local minima depth and shape characteristics information, and calculated the shape distance image according to these shape markers. Then, we imposed these shape markers on the shape distance image as its local minima. Finally, we applied the watershed algorithm again on the modified shape distance image to sepa rate the overlapped spots accurately. The experiments on the synthetic gel image and the different types of real gel images showed that compared with other improved watershed algorithm, the proposed one had a higher correct sepa- ration rate with an error less than 6%, and better separation results of overlapped spots.
