摘  要:目的用大生化速率法、干化学法与丙酮酸氧化酶法测定血清丙氨酸氨基转移酶(ALT)活性,进行比较,并对其可行性及适用性进行探讨。方法用BS-300全自动生化分析仪、Mission^TM C100干式生化分析仪和丙酮酸氧化酶法同时检测血清ALT,并对结果进行统计分析。结果精密度测定:干式法测定的ALT的变异系数(CV)值为5.58%,丙酮酸氧化酶法和大生化速率法cV值分别为4.76%和4.55%。干式法、丙酮酸氧化酶法与大生化速率法的相关系数(r)分别为0.986、0.994。根据实验结果建立了本实验室ALT正常值参考范围为0~70U/L。结论干式法和丙酮酸氧化酶法检测结果准确、快速、方便,与大生化速率法线形相关密切,均无统计学差异(P〉0.05)。使用丙酮酸氧化酶法ALT检测试剂盒可进行大批量ALT筛查,该方法具有快速、简便、精密度高的优点,特别适用于标本量大手工操作的实验室,可将阳性标本全部检出,无阳性漏检。Objective Determination of serum alanine aminotransferase activity (ALT) using biochemical rate method, dry chemistry method and pyruvate oxidase method, were compared, and to its applicability and feasibility is discussed. Methods the detection of serum ALT by BS-300 automatic biochemical analyzer, Mission^TMC100 biochemical analyzer and pyruvate oxidase method, and the results were statistically analyzed. Results the precision determination of coefficient of variation: dry method for the determination of ALT (CV) =5.58 %, pyruvate oxidase method and biochemical rate CV values were 4.76%and 4.55%. The correlation coefficient of dry method, pyruvate oxidase method and biochemical method (R) were 0.986, 0.994. But there are false positive results of pyruvate oxidase method. Conclusion dry method and pyruvate oxidase method detection result is accurate, fast, convenient, and biochemical rate normal shape related closely, there was no significant difference (P〉0.05). Kit using pyruvate oxidase method ALT for mass screening for ALT, this method has the advantages of rapid, simple, high precision, especially suitable for a large number of samples of manual laboratory, can be detected in all samples, no positive detection.
