摘 要:利用小麦成熟胚脱分化基因芯片对脱分化过程中MADS-box家族基因的表达变化进行研究,检测到MADS-box家族转录因子基因及其靶基因共19个,上调表达基因3个,下调表达基因12个,混合表达基因4个,其中,CA730918、CK213813、BE417035、BJ276784、CA726603可能在小麦脱分化过程中意义重大。利用半定量RT-PCR技术对CA670406、CK213813、CA679889、AB107992和BJ228622基因的表达变化进行验证,结果与基因芯片检测结果相似。Expression changes of MADS - box family genes during dedifferentiation were studied using mature wheat embryo dedifferentiation gene chip. Nineteen transcription factor genes and target genes of MADS - box family were detected. The number of up - regulated, down - regulated and mixed expressed genes were 3, 12 and 4 respectively. Among these, CA730918, CK213813, BE417035, BJ276784 and CA726603 may play significant roles during dedifferentiation of mature wheat embryo. The expression changes of 5 genes including CA670406, CA670406, CA679889, AB107992 and BJ228622 were verified by semi - quantitative RT - PCR technique, and its results were similar to those of gene chip detection.
