桑青枯雷尔氏菌MR111的腺苷酸激酶adk基因的克隆和序列分析

摘　　要：腺苷酸激酶是广泛存在的一种重要的核苷酸激酶。进行PCR扩增和克隆测序获得桑青枯雷尔氏菌MR111的腺苷酸激酶adk基因长502 bp的序列,NCBI在线BLAST分析其与大肠杆菌、流感嗜血杆菌同源基因的相似性在80%以上,序列编码蛋白有腺苷酸激酶保守的AMP结合结构域和LID结构域,为长型腺苷酸激酶。聚类分析结果表明,该基因与其他细菌的同源基因聚合在同一分支。Adenylate kinase is an important nucleotide kinase, and widely existed in various biological cells. Adk gene of Ralstonia solanacearum MRlll with the length of 502 bp was obtained by PCR amplication, cloning and sequencing. The similarities by NCBI online Blast between this adk gene and the homologous gene from Escherichia coli, Haemophilus influenzae, et al were above 80%. Adk of MR111 was long adenylate kinase, and had conservative AMP binding domain. Cluster aralysis showed that, adk gere and homologovs genes from other bacterium were in the same branch.
