摘 要:[目的]研究新疆紫草不同材料的染色体制片方法,为今后倍性鉴定奠定技术基础.[方法]以新疆紫草根尖及毛状根根尖为试验材料,采用常规制片法,分别考察培养方式及培养时间、取材时间、预处理方法、酸解方法、酸解时间、染色时间对染色体制片效果的影响.[结果]B5固体培养基培养4d的毛状根中期分裂相指数较大,且染色体清晰,制片效果较好;早上08:00取材的毛状根根尖的中期分裂相指数(124.2‰)最高,原植物根尖(115.4‰)在早上10:00取材较为适宜;毛状根根尖采用0.1%秋水仙素、温度15℃、预处理时间4h时的中期分裂指数为97.6‰~124.2‰,室温酸解法、10%盐酸、酸解时间为12 min、染色时间为20 min有利于毛状根根尖的制片.原植物根尖采用0.1%~0.2%秋水仙素、温度4 ~15℃、预处理时间3h、室温酸解法、10%盐酸、酸解时间为15 min时的中期分裂指数为128.6‰~128.9‰.[结论]材料不同则取材时间及秋水仙素预处理时间就有所不同.秋水仙素预处理时间、温度及酸解时间是影响新疆紫草染色体制片效果的关键因素.[ Objective ] This study aims to explore the chromosome squashing method of different materials of Arnebia Euchroma (Royle) Johnst, and provide reference for the polyploid identification. [ Method ] The roots and hairy roots of Arnebia Euchroma (Royle) Johnst's were used as experiment materials and conventional tablet method was used to investigate the influence of chromosomes squashing effects of cultivation mode, culti- vation time, sampling time, pretreatment method, acid solution method, acid solution time and dyeing time. [ Result]4d medium -term phase splitting index of B5 solid medium culture hairy roots was larger, the chro- mosomes were clear and squashing effect was good ;Sampled at 8:00 A.M. medium -term phase splitting in- dex of hairy roots apices was the highest ( 124.2%0), the roots (115.4%o) of original plant sampled at 10:00 A.M. were relatively appropriate; The medium - term phase splitting index of hairy roots apices of was 97.6%0 to 124.2%o when 0.1% colchicine solution was pretreated at 15℃ for 4h ; Acid solution at room tem- perature, hydrolysed with 10% HCL for 12 rains and dyed 20 mins was advantageous to the roots. Medium - term phase splitting index of original plant roots apices of was 128.6%o 0. 1% to 0.2% colchicine solution was pretreated at 4℃ to 15℃ for 3h and acid solution at squashing of hairy to 128.9%o when room temperaturewith 10% HCL for 15 mins. [ Conclusion] When the material was different, solution time were different, too.
