摘  要:根据转基因大豆A5547-127品系外源插入片段与植物基因组序列设计品系特异性引物,筛选最佳引物并对反应体系和反应条件进行优化,最终建立了转基因大豆A5547-127品系成分特异性LAMP的快速检测方法。对该方法进行了特异性、灵敏度和稳定性测试。结果表明:该方法能快速、特异性检测出转基因大豆A5547-127品系成分,定性检测低限为0.5%,稳定性和特异性均能达到100%。According to exogenous insert sequences connection with the plant genome sequences of transgenic soybean A5547-127, several event specific primer sets were designed. The optimum was selected from these candidates, then the reaction system and reaction conditions were optimized. Finally a loop-mediated isothermal amplification (LAMP) method for rapidly detecting transgenic soybean A5547-127 component was developed. The performances, including specificity, sensitivity and stability were tested with this method. The results showed that the method can specifically detect transgenic soybean A5547-127 component rapidly, with limit of detection (LOD) as low as 0.5 %, stability and specificity both of 100 %.
