摘 要:目的研究淫羊藿总黄酮(TFE)干预后,对模拟高原缺氧大鼠相关指标的影响,探索其预防高原病的作用机理。方法将大鼠分为6组:平原对照组、高原对照组、TFE高、中、低剂量组(710、305、150 mg.kg-1)和地塞米松组(0.5 mg.kg-1)。灌胃给药,每天1次,共给药7 d,地塞米松组给药3 d;然后放入低压氧舱,模拟大鼠高原海拔5 km,低氧暴露72 h,测定血清和脑、肺、心组织中SOD、MDA、TNOS、iNOS、NO含量水平;测定血清、脑组织的S100B及CORT含量水平;取脑、肺组织,测定其含水量。结果 TFE能够增高缺氧大鼠血清、脑组织中CORT、SOD、TNOS、NO含量,增高低压低氧大鼠肺,心组织的SOD、TNOS、NO含量水平,尤其是脑,心组织中SOD显著增高,MDA显著降低,其中以TFE高剂量效果最好。地塞米松增强缺氧大鼠血清及脑,心中CORT,SOD,TNOS,NO含量,降低血清及脑组织中S100B,MDA含量。与高原对照组比较,地塞米松组脑含水量和肺含水量有所降低,但均无显著性差异。TFE高,中,低剂量组脑含水量和肺含水量无显著变化。结论TFE对缺氧时大鼠机体损伤有保护作用,其高剂量给药组(710 mg.kg-1)效果最显著,能够不同程度提高大鼠血清、脑、肺,心组织中SOD、NOS、NO水平,降低其MDA水平,提示TFE在抗缺氧方面有良好的开发前景。Objective To study the effect of total flavonoids extract of epimedium(TFE) on hypoxic rats exposed to high altitude and to explore its role in prevention of altitude sickness.Methods The rats were divided into six groups:the plains control group,plateau control group,TFE high dose group(710 mg · kg-1),TFE middle dose group(305 mg · kg-1),TFE low-dose group(150 mg · kg-1) group and dexamethasone group(0.5 mg · kg-1).The first five groups were intragastrically administered for 7 days,once a day,while dexamethasone group was administered for 3 days.All rats were exposed to a 5 km hypoxic chamber for 72 h.The levels of SOD,MDA,TNOS,iNOS,NO in the serum,brain and lung tissues were determined.The concentration of S100B and CORT in serum and brain tissues was determined.The water content of lung and brain tissues was also determined.Results The levels of CORT,SOD,TNOS,NO in serum,brain,lung and heart tissues were increased but the levels of MDA were decreased in TFE group.In dexamethasone group,the concentration of CORT,SOD,TNOS,NO in the serum,brain and heart was incresed but the level of S100B and MDA was decreased in the serum and brain.Compared with plateau group,the brain and lung water content was slightly decreased in dexamethasone group,but no significant changes were observed in TFE groups.Conclusion TFE can improve the level of free radicals in different degrees,suggesting good prospects for the development of anti-hypoxia drugs.
