摘 要:【目的】探寻影响兔肉中脂肪酸性状的优势基因型。【方法】利用PCR-RFLP技术检测60只美系獭兔、51只伊拉肉兔FABP4基因的单核苷酸多态性,分析其与兔肉中脂肪酸含量的相关性。【结果】2个兔品种在intron1中存在C/T突变,该突变位点能被TaiⅠ限制性酶识别,且经酶切后发现,2个兔群体中存在CC、CT和TT 3种基因型。美系獭兔的3种基因型频率分别为0.083,0.467和0.450,等位基因C和T的基因频率分别为0.683和0.317;伊拉肉兔的3种基因型频率分别为0.137,0.549和0.314,等位基因C和T的基因频率分别为0.588和0.412。关联分析结果表明,在美系獭兔群体中,CC型个体肌肉组织中的脂肪酸C14:0含量和脂肪酸C18:1,n-9含量显著高于CT和TT型个体;TT型个体脂肪酸C18:3,n-3含量显著高于CC和CT型个体;TT型个体脂肪酸C20:4,n-6含量显著高于CC型个体。在伊拉肉兔群体中,CC基因型个体肌肉组织中的C18:1,n-9含量显著高于CT和TT基因型个体(P<0.05);CC型个体脂肪酸C18:2,n-6含量显著高于CT型个体;TT型个体脂肪酸C18:3,n-3含量显著高于CC和CT型个体;CT型个体脂肪酸C20:4,n-6含量显著高于CC型个体。【结论】FABP4基因可作为美系獭兔和伊拉肉兔脂肪酸性状选育的候选基因。[Objective] This study aimed to find the dominant genotype influencing fatty acid character in rabbit meat. [Method] FABP4 gene single nucleotide polymorphism of 60 Rex rabbits and 51 IRA rab- bits was detected with PCR-RFLP,and the correlation between SNP and fatty acid content of the two rab- bits was analyzed. [Result] The intronl C/T mutation existed in both rabbit breeds, which can be recog- nized by Tai I restriction enzymes. There were three genotypes: CC,CT,and TT. The genotype frequen- cies of Rex were 0. 083,0. 467 and 0. 450,and the allele gene frequencies of C and T were 0. 683 and 0. 317. The genotype frequencies of IRA were 0. 137,0. 549 and 0. 314,and the allele gene frequencies of C and T were 0. 588 and 0. 412. Correlation analysis showed that for Rex rabbits,the content of fatty acid C14:0 and C18.1,n-9 in CC genotype was significantly higher than that of CT and TT genotypes (P〈0.05);C18:3,n-3 contents of CC and CT were significantly lower than that of TT genotype~ and C20.4, n-6 content of TT genotype was significantly higher than that of CC. For IRA rabbits group, the fatty acid C18.1, n-9 content of CC genotype was significant higher than that of CC and CT genotypes (P〈0.05);C18.2, n-6 content of CC genotype was significantly higher than that of CT genotype C18.3 ,n-3 content of TT geno- type was significantly higher than that of CT and CC genotypes;and C20:4, n-6 content of CT genotype was significantly higher than that of CC genotype. [Conclusion] FABP4 gene can be used as candidate gene for fatty acids character of Rex and IRA rabbits.
