虎奶菇菌核多糖SHNP的分离纯化及形貌观测

摘　　要：利用DEAE-52纤维素柱层析和Sephadex G-1500凝胶柱层分离纯化热水浸提的虎奶菇菌核多糖,得到组分SHNP。利用紫外分光光度计和高效液相色谱鉴定其纯度,高效液相色谱测其分子量,气相色谱法进行单糖组成分析,环境扫描电镜观测了其分子形貌。结果显示,SHNP的分子量为1.463×106Da,为只含葡萄糖的大分子葡聚糖,环境扫描电镜显示SHNP固体聚集体表面形貌较为规则、平整光滑且呈现排列紧密的片状和杆柱状结构。Polysaccharide SHNP is extracted from Pleurotus tuber-regium by hot water extraction,it is purificated by DEAE-52 cellulose column and Sephadex G-150 gel filtration chromatography to obtain a component SHNP.Its purity identifies by ultraviolet spectroscopy and highperformance liquid chromatography,the molecular weight is determined by HPLC,the monosaccharide composition is determined by Gas chromatography,the molecular morphology is observed by Environmental ScanningElectron Microscope（SEM）.The results showthat the average molecular weight ofSHNP is 1.463×106Da,it is glucan composed by glucose,The surface morphology of SHNP solid aggregates is more regular,smooth and presented closely spaced sheet and rod columnar structure.
